The Challenge: Precision Engineering for Patient-Friendly Drug Delivery

GLP-1 receptor agonists, widely used for diabetes and obesity management, traditionally require injections, which can be uncomfortable for patients. Researchers at the Technical University of Denmark and Novo Nordisk aimed to develop buccal filmsβ€”thin, dissolvable films that adhere to the cheek and release medication directly into the bloodstream. However, creating these films required precise, uniform layers for controlled drug release and strong adhesion, along with scalability for consistent production.

To address these challenges, the team utilized infinityPV’s Slot-die Coater. This advanced machine enabled the precise application of multiple layers, ensuring uniformity in the mucoadhesive layer, active drug layer, and protective backing. The Slot-die Coater facilitated the production of dual-layer and triple-layer films, with the triple-layer design allowing for sequential drug release: a penetration enhancer first, followed by the GLP-1 drug. This approach improved medication absorption while minimizing irritation.

The machine’s scalability ensured consistent production, making it suitable for both research and potential commercialization. This precision and scalability were critical in overcoming the limitations of traditional manufacturing methods.

The Results: Effective, Comfortable, and Scalable Drug Delivery

Testing demonstrated that the films produced with the Slot-die Coater performed exceptionally well. The triple-layer films showed more effective drug release compared to dual-layer films and adhered securely to the buccal mucosa while preserving tissue integrity. Evaluations using porcine buccal tissue confirmed improved medication absorption and no tissue damage, indicating that buccal films could serve as a viable alternative to injections.

This breakthrough offers a pain-free, patient-friendly method for administering diabetes and obesity medications. The success of this project highlights the potential for more patient-friendly treatments in the future, with tools like the Slot-die Coater playing a crucial role in advancing medical innovation.
